The visitors continued to accumulate runs at will before declaring at a mammoth 575/6.
Their bowlers then made quick work of the Bangladesh lineup, leaving the hosts in disarray at 38/4 by the close of play.
South Africa resumed their dominance after the break, with David Bedingham taking charge of the scoring.
He struck a flurry of boundaries, before Tony de Zorzi followed suit with successive fours to push past 150.
The runs kept flowing even after the drinks break until Taijul Islam finally made a breakthrough.
The left-arm spinner bowled Bedingham out after he had passed fifty, and Bangladesh capitalized on the opening, claiming two more wickets in quick succession to enter the lunch break on a high.
The hosts kept the pressure on after lunch, taking the last four South African wickets for just 37 runs.
However, any hopes of Bangladesh keeping South Africa under 475 quickly faded as Wiaan Mulder and Senuran Muthusamy took charge.
Mulder continued his excellent form, bringing up another half-century, while Muthusamy made a rapid start.
Together, they added a century stand, with Muthusamy bringing up his own fifty shortly after the break in the final session.
At one point, it seemed Bangladesh might be on the verge of a breakthrough, but Mulder dashed any hopes by launching Taijul Islam straight down the ground for a six, bringing up his maiden Test century and prompting the declaration.
With a monumental chase ahead of them, Bangladesh were immediately on the back foot in their reply.
Kagiso Rabada, newly crowned World No. 1, struck with the very first ball of his spell, getting Shadman Islam caught down the leg side for a duck.
Zakir Hasan followed shortly after, edging a delivery to the keeper.
Dane Paterson and Keshav Maharaj then got in on the act, bagging wickets in successive overs, reducing Bangladesh to 32/4 after just seven overs.
Bad light then brought an early end to their misery, with the players heading off the field, just as they had on day one.
